Hi Twan,
The subtitle option is available only for text box objects.
Charts offer a single title field only. You can create two-line titles (using a line break), but the lines will share the samme formatting.
I would probably experiment with adding a label or textbox object on top of the chart to give the visual impression of a subtitle.

If you would like to have subtitles in another font or size then the main title, I think it's not possible.
But you can make a title with dynamic content or add space between title and subtitle, like in this screenshot:

I mean that you cannot make a subtitle with another font. We do that now with a text box, but is not ideal. For us it would be a nice feather if this is possible.
A funny coincidence is that I also had the customer request today if two different colors can be used in a chart headline. That might be a nice feature request :)
